Technology Express---Research on the Molecular Mechanism of Anti-cancer Drugs in JAK/Stat Signal Pathway by OpenSPR

Cervical cancer is the second most common malignant tumor in women. In recent years, its incidence has become younger. The isomeric compounds TAD-1822-7-F2 (F2) and TAD-1822-7-F5 (F5) designed based on the chemical structure of taspine and its derivatives can effectively inhibit the proliferation of HeLa cells. Researchers at Xi'an Jiaotong University used OpenSPR's surface plasmon resonance (SPR) technology to delve into the intrinsic mechanisms by which F2 and F5 inhibit Hela cell proliferation. The study "Novel compounds TAD-1822-7-F2 and F5 inhibited HeLa cells growth through the JAK/Stat signaling pathway" was published in "Biomedicine & Pharmacotherapy" in May 2018.

The JAK/STAT pathway is closely related to the development of tumors. Previous studies have found that F2 and F5 inhibit the proliferation of Hela cells. To investigate whether the inhibitory mechanisms of F2 and F5 are related to JAK2 protein, this study used Nicoya's OpenSPR to detect the interaction between F2/F5 and JAK2. The protein was immobilized on the NTA chip, and then different concentrations of F2/F5 were passed through the chip, and the kinetic parameters were analyzed. It was found that both F2/F5 and JAK2 have strong affinity, and the molecular target of F2/F5 inhibiting cancer cell proliferation was quickly determined.

SPR is a label-free detection technique that quantifies the binding between two molecules and determines the rate constant, the dissociation rate constant, and the affinity of the intermolecular interactions. Compared with the traditional ELISA, pull-down and other end-point detection techniques, SPR technology has the advantages of simple operation, real-time observation and rich data.
